Pacific Biosciences
One of the newest additions to our portfolio is the PacBio Sequencing service. Pacific Biosciences developped a third generation DNA sequencer that provides real time analysis of biomolecules with single molecule resolution.
Key advantages of the PacBio RS:
- Very long read lengths
- High single molecule and consensus accuracy
- Balanced coverage & minimal GC-bias
- Unique kinetic information (base modification detection)
SMRT sequencing technology:
One of the major features of this sequencer is the ability to produce very long reads. Only a small amount of organisms today have been completely sequenced, due to the limitations of second generation sequencers. Examples of these challenges are the inability to sequence GC-rich regions, repeats or solve structural variation with a high confidence. PacBio RS is able to tackle these problems due to the generation of ultra long reads, the high accuracy that can be obtained via circular consensus sequencing (CCS), and the minimal GC-bias providing a balanced coverage. Applications specific for PacBio sequencing:
- Multi-Molecule consensus:
- Microbial de novo assembly
- BAC de novo accembly
- Gap closing or scaffolding
- Structural variation detection
- Amplicon sequencing
- Gene-panel sequencing
- Haplotype phasing
- Base-modification detection
- Single-Molecule sonsensus
- Viral quasi-species
- Rare resistance mutations
- Rare, closely related transcript isoforms
- Low-frequency heteroplasmy
- 16s metagenomics
- DNA-damage detection
Complete genome assemblies:
The combination of ultra long read generation and the understanding of random error occurence (rather than biased errors of other sequencing platforms), you are able to completely finish genomes with high-quality and accuracy.
Accurate characterization of large structural variations:
Due to the generation of long read lengths, PacBio RS can easily sequence large repeat regions and resolve complex structures.
Unbiased genome coverage:
The PacBio RS provides balanced coverage and minimal GC-bias that is needed for high-quality assembly of high or low GC content organisms or regions. The combination of minimal GC-bias and the generation of long reads, enables the sequencing of large GC repeat regions with high accuracy.
Hybrid assembly:
PacBio sequencing is ideal in combination with other established technologies such as Illumina Hiseq2000 to assemble genomes. These second generation sequencers provide shorter reads with a higher accuracy, while the PacBio RS is able to generate ultra long reads reducing the number of contigs. Using the long reads and the random error occurence, you can solve complex large structural variations or repeat regions.
